OTC Sleep Aids (Diphenhydramine-Based)
Many over-the-counter sleep aids use diphenhydramine as an active ingredient — the same compound in Benadryl. Diphenhydramine is a known anticholinergic, meaning it interacts with acetylcholine signaling. Acetylcholine is the primary neurotransmitter for memory encoding and recall. The Gray et al. JAMA Internal Medicine study found cumulative anticholinergic drug use associated with a 54% higher dementia-risk association, with the strongest association in individuals who had used these medications for more than 3 years total.

Beta-Blockers (Especially Propranolol)
Propranolol — the lipophilic beta-blocker used for hypertension, performance anxiety, and migraines — is discussed for blood-brain-barrier penetration and norepinephrine-signaling context.
